▶ Storage Buffer
Approximately 0.1 - 0.4 mg/ml of each proteins are in the buffer (20 mM Tris phosphate, pH 7.5 at 25°C), 2% SDS, 0.2 mM Dithiothreitol, 3.6 M Urea, and 15% (v/v) Glycerol.
▶ Storage condition
Can be stored maximum 3 months at 4°C, For long term storage, store at -20°C.
Please divide the products in small volumes and store them separately to avoid repetitive freezing and thawing.
▶ Quality control assay
Our SDS-Polyacrylamide gel anlaysis clearly shows distinguighable 12 bands.
▶ How to use
- Thaw the ladder either at room temperature or at around 37 - 40°C for a few minutes to dissolve the precipitated solids (Do not boil).
- Mix thoroughly to ensure homogeneity.
- Load the following volumes of the ladder on the SDS-PAGE gel:
- 5 μl/well for mini-gels, 2.5 μl/well for blots
- 10 μl/well for large-gels, 5 μl/well for blots
- Load the samples.
- Connect the power supply and start the electrophoresis.
▶ Guide for Estimating Molecular Weight Estimation (kDa)
The migration patterns of AccuLadder™ 3-color Prestained Protein size marker (Broad) for different electrophoresis conditions can be viewed in the diagram below. In the prestained protein marker, the coupling of the protein chromophores is affected more in the apparent molecular weights in the SDS-PAGE compared to that of the unstained ones. Therefore, to achieve the accuracy of protein molecular weight measurements to be more than 95%, we recommend using the unstained protein markers.
