AccuYeast™ Build Strain

AccuYeast™ Build Strain is a customized service supporting both yeast genome integration and episomal plasmid engineering. For genome integration, gene editing options include knock-in, knock-out, and knock-down. In addition, episomal plasmid engineering enables the construction of plasmids that are maintained and expressed in yeast cells.

Genome integration involves inserting foreign DNA into a specific locus of the yeast genome or deleting/modifying an existing gene. In contrast, episomal plasmid engineering enables gene expression using plasmids that replicate independently within yeast cells without genomic insertion. The appropriate option can be selected depending on research objectives and experimental design.

  • Knock-in: Insertion of a specific gene into a targeted locus of the yeast genome
  • Knock-out: Deletion of a target gene to eliminate its function
  • Knock-down: Reduction of gene expression levels

The desired editing strategy can be specified in the Desired Editing Type section of the Order Form.

This service is performed using customer-provided yeast strains. Genome integration or plasmid engineering is conducted based on the supplied strain. Detailed instructions for strain preparation and shipping are provided during the ordering process.

For genome integration projects, customers specify the target locus within the yeast genome. Homology arms (HAs) are used to mediate homologous recombination at the target site. Genome-based HA sequences are provided by default, and customized sequences can be arranged upon consultation if required.

Customers may provide their own DNA sequences or, when needed, integrate BIONEER’s Gene Synthesis services to proceed seamlessly from sequence design to yeast strain construction. Gene Synthesis is optional and can be selected in the Gene Synthesis Option section of the Order Form.

DNA cassettes are designed according to the purpose of genome integration or plasmid engineering. For genome integration, the cassette may include left and right homology arms, ORF(s), promoter, terminator, and selection marker. The total cassette size (bp) must be specified in the Order Form. Customers may provide the full cassette sequence or define individual components only.

All engineered yeast strains undergo colony PCR and Sanger sequencing as standard QC to confirm correct insertion or editing. For large inserts or projects requiring structural confirmation, NGS-based analysis may be applied for additional validation. The QC scope is defined in advance based on the order details and cassette design.

The completed yeast strain is delivered in one of the following storage formats:

  • Agar type: Yeast strains are cultured on nutrient-rich solid medium and stored at 4 °C to suppress further growth prior to delivery.
  • Glycerol type: Liquid-cultured yeast strains are prepared with 30% (v/v) glycerol for long-term storage and can be preserved at −70 °C.

The delivery formats are as follows, and customers may select their preferred option during ordering:

  • Agar type: Rich medium + supplements, solid medium (2.0 mL microtube)
  • Glycerol type: Rich medium + supplements + 30% glycerol, liquid medium (2.0 mL microtube)

AccuYeast™ Build Strain is a yeast engineering service powered by BIONEER's gene synthesis expertise, offering precise genome integration and episomal plasmid engineering. The service can be seamlessly integrated with BIONEER's Gene Synthesis service, enabling support from the design stage through strain construction. Based on the provided DNA cassette, engineered yeast strains are delivered as QC-validated yeast strains, ensuring reliable performance and quality.
